A stirred microreactor monitored by spectrophotometry,was attached via flow through port to the LOV module and used to study kinetics of acetylcholine estherase in absence and presence of inhibitors.
Enzyme, substrate and inhibitor were precisely metered by a syringe pump (A) into the holding coil and transferred rapidly into the microreactor. The thus prepared homogeneous mixture of reactants of known concentrations was monitored during the initial reaction rate in absence (A) and presence (B) of inhibitors as well as in absence of enzyme (blank). The collected data allowed to determine Michaelis constants for the reactions studied, and to identify the type of inhibition (competitive, non-competitive, mixed). The auxiliary pump (B) was used to empty and flush the micro reactor.
